Resumen del Seminario:

Why can some tumor cells survive despite an active antitumor T-cell response while others die?

Building on nearly a decade of work on tumor-intrinsic resistance, I will show how we use three complementary approaches to address this question. Genome-scale CRISPR screening provides an unbiased strategy to identify resistance determinants, sensitizers and tumor vulnerabilities, while drug and combination screening provides a translational path to identify synergistic combinations that enhance immunotherapy efficacy and overcome resistance. We have also established a quantitative live-cell imaging methodology to study the immune synapse, allowing us to follow tumor–T-cell interactions over time and evaluate how genetic or pharmacological perturbations alter contact dynamics and killing.

Using solid and hematological cancer models treated with CAR-T cells, T-cell engagers or immune checkpoint blockade, I will discuss how these approaches, together with mechanistic validation, can translate screening hits and functional phenotypes into biological insight and help identify biomarkers, therapeutic vulnerabilities and rational combination strategies.

Beyond our own research questions, I will show how these methodologies are being applied in ongoing collaborationsand could support new projects within CIBERONC and the broader immuno-oncology community.

Short Bio:

Dr Enrique J. Arenas is a translational cancer researcher with more than 15 years of experience across IRB Barcelona, VHIO and the Josep Carreras Leukaemia Research Institute, currently based at the Sant Pau Research Institute within Dr. Manel Esteller’s laboratory. His research has focused on cancer immunotherapy and tumor-intrinsic resistance for nearly a decade. Since 2023, he has led an independent research line aimed at understanding and overcoming resistance to T-cell–based immunotherapies.
